ANTIOXIDANT EFFECTS OF Convolvulus pluricalus IN RAT BRAIN MITOCHONDRIA AGAINST OXIDATIVE DAMAGE INDUCED BY GAMMA RADIATION AND PHOTOSENSITIZATION

چکیده
Aqueous and methanolic extracts of natural herb, Convolvulus pluricalus (CP) was monitored for their antioxidant ability against photosensitization and radiation induced damages in rat brain mitochondria. Both the extracts of CP (50 μg /ml assay) showed significant protection against the formation of the lipid peroxidation product, TBARS. Protein oxidation and degradation of proteins was shown to be significantly high following photosensitization, while CP extracts could effectively modify these damages. Exposure of plasmid DNA pBR322 to gamma radiation resulted in enhanced formation of single strand breaks (SSB). Simultaneous treatment of pBR322 DNA with CP extracts during radiation effectively prevented the damage. Further, these extracts also showed inhibition against stable radicals, 2, 2’azino-bis (3ethylbenzthiazoline-6sulfonic acid (ABTS) and 1, 1diphenyl-2-picryl hydrazyl (DPPH). The scavenging capacity of the extracts was quantified by pulse radiolysis experiments. Hence, Convolvulus pluricalus may emerge as potent antioxidant/radioprotector, capable of scavenging singlet oxygen, hydroxyl radical, as well as other stable radicals, ABTS and DPPH.

Melanin acts as a potent UVB photosensitizer to cause an atypical mode of cell death in murine skin.
Melanin protects the skin against DNA damage induced by direct absorption of sunlight's UV radiation. Yet, irradiating melanin in vitro or in cultured cells also generates active oxygen species such as superoxide, which can indirectly induce oxidative base lesions and DNA strand breaks. This photosensitization is greater for pheomelanin (yellow and red melanin) than for eumelanin (brown and bla...
متن کاملThe role and mechanisms of zinc oxide nanoparticles in the improvement of the radiosensitivity of lung cancer cells in clinically relevant megavoltage radiation energies in-vitro
Objective(s): Semiconductor zinc oxide nanoparticles (ZnO NPs) have unique properties, such as inherent selectivity and photosensitization effects under ultraviolet (UV) radiation. ZnO NPs serve as promising anticancer agents. However, UV radiation limits their penetration into the body. In most clinical settings, it is essential to use high-energy photons in the treatment of deep-seated tumors...
متن کامل3-hydroxypyridine chromophores are endogenous sensitizers of photooxidative stress in human skin cells.
Photocarcinogenesis and photoaging are established consequences of chronic exposure of human skin to solar irradiation. Accumulating evidence supports a causative involvement of UVA irradiation in skin photo-damage. UVA photodamage has been attributed to photosensitization by endogenous skin chromophores leading to the formation of reactive oxygen species and organic free radicals as key mediat...
متن کامل5-Methyltetrahydrofolate inhibits photosensitization reactions and strand breaks in DNA.
The known functions of folate are to support one-carbon metabolism and to serve as photoreceptors for cryptochromes and photolyases. We demonstrate that 5-methyltetrahydrofolate (5-MTHF, the predominant folate in plasma) is also a potent, near diffusion limited, scavenger of singlet oxygen and quencher of excited photosensitizers. Both pathways result in decomposition of 5-MTHF, although ascorb...
متن کاملMelanin Photosensitization and the Effect of Visible Light on Epithelial Cells
Protecting human skin from sun exposure is a complex issue that involves unclear aspects of the interaction between light and tissue. A persistent misconception is that visible light is safe for the skin, although several lines of evidence suggest otherwise. Here, we show that visible light can damage melanocytes through melanin photosensitization and singlet oxygen (1O2) generation, thus decre...
